The Novodiax ihc Magenta 1:1 Kit—which consists of a ready-to-use magenta chromogen dye and an accompanying diluent—is intended for use as part of immunohistochemistry (IHC), immunocytochemistry (ICC), and with other immunoassays that utilize horseradish peroxidase (HRP) or polymerized HRP (pHRP) detection systems. The magenta chromogen substrate is optimized for use on both frozen tissue and formalin-fixed paraffin-embedded (FFPE) tissue.
Principles of the procedure
When performing an IHC assay, antibodies are applied to frozen or FFPE tissue sections where they bind to the matching antigen if it is present. After a thorough washing step, the Magenta 1:1 working solution may then be applied to the slide. The antibody-linked HRP catalyzes the chromogen substrate to form a visible magenta-colored (reddish-purple) product at the site of the antibody-antigen reaction. The specimen may subsequently be further processed and counterstained with hematoxylin, after which mounting media and/or a coverslip can be applied. Results are to be viewed and interpreted using a light microscope.
Benefits of the Magenta Chromogen
- Compared to DAB chromogen, ihc Magenta 1:1 allows users to better differentiate positive staining from natural brown pigments (e.g. melanin in skin tissue, and hemosiderin which is sometimes found in the lungs, liver, and kidney under disease conditions).
- User friendly 1:1 reagent format where chromogen substrate is prepared by simply mixing equal parts of the Magenta Chromogen and Magenta Diluent.
- Chromogen working solution remains stable for up to seven (7) days after mixing.
- Permanent record—proprietary chemistry allows ihc Magenta 1:1 stains to withstand dehydration and clearing with alcohol and xylene, so they are compatible with both aqueous and organic mounting media.
